Problems related to the lack of functional swimbladder in intensive rearing of the seabass Dicentrarchus labrax and the sea bream Sparus auratus . ArchiMer
Chatain, Beatrice.
The primordial inflation of swimbladder normally occurs at a size of 4 to 5 mm in sea bream Sparus auratus and 5 to 6 mm in sea bass Dicentrarchus labrax . However, in intensive larval rearing conditions, lack of inflation is often observed and leads to the production of up to 80% of postlarvae whose swimbladder is not functional. They are, on the average, 20-30% smaller in weight than normal fry. Their survival rate can also be reduced by any kind of stress such as weaning, handling or hypoxic conditions. It is suggested that the selective mortality which affects the abnormal fish results both from a decrease in predatory efficiency and an increase in energetic needs.

Estimation et amélioration des performances zootechniques de l'élevage larvaire de Dicentrarchus labrax et de Sparus auratus ArchiMer
Chatain, Beatrice.
By gathering specific measurement criteria and an objective analysis of field methods, the main purpose of this work is to help estimate larval Sea Bass (Dicentrarchus labrax) and Sea Bream {Sparus auratus) stock performance raised under intensive conditions. As such, it is a methodology guide zootechnicians can use, in their daily quantitative and qualitative evaluation stock issues. The first part of this work covers defining the precision of current counting methods and establishing their limitations. The quantification methods analysed, as well as the new proposed methods, relate to the major phases of larval farming, from laying to postlarval stage. Three field methods commonly used in aquaculture to quantify egg population are therefore...

Genetical improvement of marine fish and shellfish : a french perspective ArchiMer
Boudry, Pierre; Chatain, Beatrice; Naciri-graven, Yamama; Lemaire, Christophe; Gerard, Andre.
In France, marine fish and shellfish farming is based mainly on the breeding of wild species, whose natural populations are traditionally exploited. Research programs are conducted by IFREMER, in co-operation with INRA and Universities. French research programs for the genetical improvement of marine fish and shellfish concern primarily seabass (Dicentrarchus labrax) and oysters (Ostrea edulis and Crassostrea gigas). The following programs are developed: •The knowledge of wild and farmed populations. The most recent prodjects involve the highly variable microsatellite markers. Seven markers are presently available for seabass and the work is now initiated on turbot (Scophthalmus maximus). In molluscs, microsatellite markers have been developed for the...

Pressure and cold shock induction of meiotic gynogenesis and triploidy in the European sea bass, Dicentrarchus labrax L.: relative efficiency of methods and parental variability ArchiMer
Peruzzi, Stefano; Chatain, Beatrice.
The optimal conditions for the retention of the second polar body in sea bass eggs were investigated by altering the timing, intensity and duration of application of pressure and cold shocks. Treatment optima for cold shocks were 0-1 degrees C for 15-20 min at 5 min after fertilisation (a.f.) and 8500 psi for 2 min at 6 min a.f. for pressure shocks. Meiogenesis was obtained by fertilising eggs with UV-irradiated homologous sperm (32,000 erg mm(-2)) and pressure or cold shocking eggs as above. 100% triploidy was induced following definition of liable periods for the disruption of the meiotic spindle obtained in gynogenesis. Ploidy investigations were performed on experimental groups by flow-cytometry. Verification of uniparental transmission in meiogens was...

Tank wall colour and light level affect growth and survival of Eurasian perch larvae (Perca fluviatilis L.) ArchiMer
Tamazouzt, Lakhdar; Chatain, Beatrice; Fontaine, Pascal.
The influence of interaction between light intensity and tank wall colours on survival and growth of perch larvae (Perca fluviatilis L.) was investigated for 15 days after hatching. Three light intensities (250, 400 and 800 Ix) and four wall colours (black, dark grey, light grey and white) were used. At the end of the experiment, survival rates ranged between 1% and 26%, and varied according to the different treatments (P = 0.0001; df = 11). A global effect of light intensity was detected(P = 0.0001; df = 2) and the highest survival rate was observed under 250 Ix. Survival was also influenced by the tank wall colour(P = 0.0001; df = 3): light grey showed the best result (17%). The greatest growth in weight and length was observed in tanks with light grey...

Impact of aquaculture on the genetic structure of Mediterranean populations of Dicentrarchus ArchiMer
Bahri Sfar, Lilia; Lemaire, Christophe; Chatain, Beatrice; Divanach, Pascal; Ben Hassine, Oum Kalthoum; Bonhomme, Francois.
A phylogeographic analysis based on 6 polymorphic microsatellites was performed on 15 sea bass samples. five from Western Mediterranean, seven from Eastern Mediterranean and three coming from French aquaculture stocks. Among the eastern samples, three did not cluster according to their geographic origin but rather with the occidental group. Furthermore. a somewhat lower allelic diversity was observed within these particular samples, indicating that they originated from a limited number of progenitors of foreign origin. Among the aquaculture stocks, only one revealed a significant reduction of genetic variability, indicating that these stocks are largely outbred and open to wild fishes. The fact that the use of occidental fingerlings to seed broodstocks in...

A shortcut method for sexing juvenile European sea bass, Dicentrarchus labrax L. ArchiMer
Menu, Bruno; Peruzzi, Stefano; Vergnet, Alain; Vidal, Marie-odile; Chatain, Beatrice.
The objective of this study was to develop a rapid and effective method of sexing juvenile sea bass with minimum labour and material. To this end, the gonad squash mount technique was applied along with macroscopic techniques for sexing a large number of experimental fish at the age of 215-275 days post fertilization (p.f.). At this age, 90% of the 3894 fish could be unambiguously sexed by macroscopic examination of their gonads, whereas the remaining proportion was identified using squash mount preparations (10%). The accuracy of the observations was measured up to classical histology procedures. Undifferentiated fish accounted for 0.1% of the total population only. The application of the squash mount technique holds out the prospect of considerable...

"Freezebass" Setting up of a sperm cryobank for sea bass. Q5CT-2002-71209. Final report (01 January 2003 – 30 June 2005) ArchiMer
Chatain, Beatrice; Poline, Olivier.
Objectives : Presently, most of marine fish farmers around the Mediterranean basin grow genetically wild fish strains, which have not been genetically improved for culture performance (growth, appearance, disease resistance etc.). It is inevitable that the present gap for genetically improved strains of marine fish, and their reliable supply, will be filled. It will ultimately dominate the competitive component of marine culture industries once they evolve into commodity-style agribusiness (with fish prices being set at the lowest possible cost of production and the best quality). Those countries, which can competitively develop and supply the improved strains, will enjoy the benefit (perhaps survival) for their local industries. They will avoid...

Flow cytometric determination of genome size in European seabass (Dicentrarchus labrax), gilthead seabream (Sparus aurata), thinlip mullet (Liza ramada), and European eel (Anguilla anguilla) ArchiMer
Peruzzi, Stefano; Chatain, Beatrice; Menu, Bruno.
The nuclear DNA content in Dicentrarchus labrax, Sparus aurata, Liza ramada, and Anguilla anguilla, was measured by flow cytometric analysis. Male human leukocytes were used as internal reference cells. Nucleated fish erythrocytes were stained simultaneously to human leukocytes with a staining buffer containing propidium iodide (PI). Nuclear DNA contents of target species were estimated in relation to an assigned value of 7.0 pg DNA for male human leukocytes. The DNA content/nucleus (+/- CI) was 1.55 +/- 0.02 pg for D. labrax, 1.90 +/- 0.03 pg for S. aurata, 1.57 +/- 0.02 pg for L. ramada, and 2.43 +/- 0.04 pg for A. anguilla. Intraspecific DNA content variations ranged from 0-9% and averaged around 4% in all taxa. The results did not evidence significant...

Haematological and physiological characteristics of diploid and triploid sea bass, Dicentrarchus labrax L. ArchiMer
Peruzzi, Stefano; Varsamos, Stamatis; Chatain, Beatrice; Fauvel, Christian; Menu, Bruno; Falguiere, Jean-claude; Severe, Armelle; Flik, Gert.
The purpose of this study was to determine whether diploid and triploid sea bass differed in terms of main haematological and physiological characteristics. Diploid and triploid fish were produced by sub-optimal pressure treatments and held in communal environments under standard rearing conditions. Total red blood cell count (RBCC), haemoglobin concentration (Hb), hematocrit (Hct), mean cell volume (MCV), mean cellular haemoglobin content (MCH), mean cell haemoglobin concentration (MCHC), plasma metabolites, osmotic pressure, gill Na+/K+-ATPase activity, electrolytes, cortisol, and 3,5,3'-triiodo-L-thyronine (T-3) were measured and compared. Triploidisation in sea bass led to an increase in erythrocyte size (32% in cytoplasm surface area, and 50% in...

Estimates of heritability and genotype-environment interactions for body weight in sea bass (Dicentrarchus labrax L.) raised under communal rearing conditions ArchiMer
Saillant, Eric; Dupont Nivet, Mathilde; Haffray, Pierrick; Chatain, Beatrice.
Genetic parameters for somatic growth rate in sea bass were estimated in two experiments where 27 (year-class 1997) to 30 (year-class 1998) families of sea bass (3 dams mated with 9-10 sires according to a full factorial design) were raised mixed in the same tanks starting shortly before hatching (48 h post fertilization). Trials were performed under two temperature (experiment 1, 1997: group high temperature (HT) and group low temperature (LT)) and two density regimes (experiment 2, 1998: group high density (HD) and group low density (LD)) with a subset of the mating design (18 families) being reproduced in the two experiments. All the fish were genotyped at 3-6 microsatellite loci which allowed them to be assigned to the individual breeders used in the...

Genetic investigation of swimbladder inflation anomalies in the European sea bass, Dicentrarchus labrax L. ArchiMer
Peruzzi, Stefano; Westgaard, Jon-ivar; Chatain, Beatrice.
Although the actiology of swimbladder inflation anomalies in important aquaculture species such as European sea bass D. labrax is not fully determined, culture conditions are commonly suggested as main contributory factors. Little information is available on whether swimbladder inflation has a genetic basis for its expression too. In this work, 24 full-sibling sea bass families from a 4 dams x 6 sires factorial crossing were reared under communal conditions. The larvae developing normal and abnormal (uninflated or hyper-inflated) swimbladders were genotyped at four microsatellite loci, Labrax-3, Labrax-13, Labrax-17, Labrax29, and allocated to the individual breeders. Out of 273 offspring, 97% could be assigned to a single parental pair. The genotype and...

A polygenic hypothesis for sex determination in the European sea bass - Dicentrarchus labrax ArchiMer
Vandeputte, Marc; Dupont Nivet, M; Chavanne, H; Chatain, Beatrice.
Polygenic sex determination, although suspected in several species, is thought to be evolutionarily unstable and has been proven in very few cases. In the European sea bass, temperature is known to influence the sex ratio. We set up a factorial mating, producing 5.893 individuals from 253 full-sib families, all reared in a single batch to avoid any between-families environmental effects. The proportion of females in the offspring was 18.3%, with a large variation between families. Interpreting sex as a threshold trait, the heritability estimate was 0.62 +/- 0.12. The observed distribution of family sex ratios was in accordance with a polygenic model or with a four-sex-factors system with environmental variance and could not be explained by any genetic...

Heritabilities and GxE interactions for growth in the European sea bass (Dicentrarchus labrax L.) using a marker-based pedigree ArchiMer
Dupont Nivet, M; Vandeputte, Marc; Vergnet, Alain; Merdy, O; Haffray, P; Chavanne, H; Chatain, Beatrice.
253 full-sib families from 33 males and 23 females of European seabass were produced in a partly factorial mating design. All fish were reared in the same tank during 14 months, then 7000 of them were dispatched in four farms to different locations (France, Israel, Italy, Portugal) representing a wide variety of environmental conditions. Around 400 g mean weight, 1177 to 1667 fish in each site were weighed and length was measured. Condition factor (K) was calculated. Pedigrees were redrawn a posteriori using microsatellites markers: parental origin could be retraced for 99.2% of fish. Due to a high incidence of deformities, the useful sample size was reduced to 491-670 fish per site. Maternal effects were small. Using a simple animal model, heritability...

Cloning and retinal expression of melatonin receptors in the European sea bass, Dicentrarchus labrax ArchiMer
Sauzet, S; Besseau, L; Perez, P; Coves, Denis; Chatain, Beatrice; Peyric, E; Boeuf, G; Munoz Cueto, J; Falcon, J.
Melatonin contributes to synchronizing behaviors and physiological functions to daily and seasonal rhythm in fish. However, no coherent vision emerges because the effects vary with the species, sex, moment of the year or sexual cycle. And, scarce information is available concerning the melatonin age, receptors, which is crucial to our understanding of the role melatonin plays. We report here the full length cloning of three different melatonin receptor subtypes in the sea bass Dicentrarchus labrax, belonging, respectively, to the MT1, MT2 and Mellc subtypes. MT1, the most abundantly expressed, was detected in the central nervous system, retina, and gills. MT2 was detected in the pituitary gland, blood cells and, to a lesser extend, in the optic tectum,...

Genetic variability in residual feed intake in rainbow trout clones and testing of indirect selection criteria ArchiMer
Grima, Laure; Quillet, E; Boujard, Thierry; Robert Granie, C; Chatain, Beatrice; Mambrini, M.
Little is known about the genetic basis of residual feed intake (RFI) variation in fish, since this trait is highly sensitive to environmental influences, and feed intake of individuals is difficult to measure accurately. The purpose of this work was (i) to assess the genetic variability of RFI estimated by an X-ray technique and (ii) to develop predictive criteria for RFI. Two predictive criteria were tested: loss of body weight during feed deprivation and compensatory growth during re-feeding. Ten heterozygous rainbow trout clones were used. Individual intake and body weight were measured three times at three-week intervals. Then, individual body weight was recorded after two cycles of a three-week feed deprivation followed by a three-week re-feeding....

Response to domestication and selection for growth in the European sea bass (Dicentrarchus labrax) in separate and mixed tanks ArchiMer
Vandeputte, Marc; Dupont Nivet, M; Haffray, P; Chavanne, H; Cenadelli, S; Parati, K; Vidal, Marie-odile; Vergnet, Alain; Chatain, Beatrice.
Selective breeding of European sea bass (Dicentrarchus labrax) receives a growing interest, as the estimated heritability of growth is medium to high. In this study, we compared the offspring of four groups of sea bass sires, mated with the same wild dams: wild (W), first generation of domestication (D), first generation of mass selection for length (M), first generation of PROSPER-like selection for length (P). The comparison was done both in replicated tanks (separate rearing) and in mixed tanks (mixed rearing) where sire origins were recovered by genotyping of eight microsatellite markers. Weight, length and growth rate were measured from day 238 post-fertilization (69 g mean weight) to day 611 post-fertilization (390 g mean weight). Both in mixed and...

Risk-taking behaviour variation over time in sea bass Dicentrarchus labrax: effects of day-night alternation, fish phenotypic characteristics and selection for growth ArchiMer
Millot, Sandie; Begout, Marie-laure; Chatain, Beatrice.
Differences in bold and shy personality on sea bass Dicentrarchus labrax were investigated between a population (wild) produced from wild-brood fish and a population (selected) produced from selected-brood fish. During the experiment (112 days), fish were reared under self-feeding condition to characterize the feeding behaviour of each individual fish. Three risk-taking tests (T1, T2 and T3 of 24 h with day-night alternation) were carried out at > 1 month intervals on 180 fish of each strain in order to monitor D. labrax behaviour over time and in relation to the light:dark period. A risk-taking score was evaluated via a preference choice between a safe zone (without food) and a risky zone (potentially with food) by recording the number and the duration...
